In this contribution the author focuses on the saprophytic phase of the life cycle of the phytopathogenic fungus Taphrina deformans (Berk.) Tul. in vitro. A method of isolation from naturally infected peach leaves (Prunus persica L. Batsch) is described. Mycelium of the isolates is characterized visually and microscopically with respect to origin, nutrient substrate and temperature. Growth and development of fungal cells depend on substrate quality. Ascus formation in vitro is probably induced by a suitable combination of nutrients. Optimal temperature for growth in vitro is 18–20 °C. Lower and higher temperatures inhibit growth.
